The effect of lipoic acid on mammalian growth is not clear, and little is known of its biosynthesis. The thioester group of lipoic acid can be reduced in various ways. In the laboratory, the oxidized lipoic acid can be reduced by strong reducing agents (zinc). The longer wavelength of ultraviolet radiation also reduces the oxidized form. In the cell, enzymes catalyze the reduction of lipoic acid. Chapter I Cellular Sources of Energy In as much as reduced lipoic acid is an antioxidant, it is difficult to distinguish a specific biological action from a nonspecific antioxidant effect.

The hydrolysis is either spontaneous or catalyzed by a specific lactonase, an enzyme which has been found in yeast, bacteria, and mammalian tissues. Although the first step in the transformation of glucose-6-phosphate to phosphogluconic acid is reversible, the second is irreversible. Glucose-6-phosphate dehydrogenase was crystallized from bovine mammary glands and from yeast. It is inhibited by certain steroids (dehydroisoandrosterone, pregnenolone), but the physiological significance of such inhibitory effects is not known.

To explain these results, the authors postulated an intermediate common to cis-aconitate and isocitrate consisting of a tricarboxylic acid fonning a complex with iron and cysteine.
